What does the IC50 value tell you?

What does the IC50 value tell you?

Half-maximal inhibitory concentration (IC50) is the most widely used and informative measure of a drug’s efficacy. It indicates how much drug is needed to inhibit a biological process by half, thus providing a measure of potency of an antagonist drug in pharmacological research.

What is a good IC50 value for a drug?

10 micromolar
In most cases, the IC50 of the best candidate compound should be lower to 10 micromolar, acceptable for NIH to screen the NCI60 program. In addition, the drug design should think over the related problem about the absorption, distribution, metabolism and excretion in our body.

What does the IC50 value tell us about how an inhibitor binds to its target protein?

IC50 is an operational parameter defined as the concentration of inhibitor (ligand) required for achieving 50\% inhibition (binding saturation) of the enzyme (receptor). The reasoning is that, in order to inhibit 50\% of the enzyme you must add, at least, the same amount (concentration) of inhibitor.

How is drug potency determined?

Potency is the concentration (EC50) or dose (ED50) of a drug required to produce 50\% of that drug’s maximal effect. Efficacy (Emax) is the maximum effect which can be expected from this drug (i.e. when this magnitude of effect is reached, increasing the dose will not produce a greater magnitude of effect)

What is efficacy of a drug?

A drug’s efficacy is a measure of the ability of the drug to treat whatever condition it is indicated for. It is not a statement about the drug’s tolerability or ease of use. Effectiveness is a measure of how well the drug works and encompasses all 3 of these issues.

Is a lower IC50 better?

The IC50 value is correlated with drug potency, i.e. the amount of drug necessary to produce the effect—the lower the IC50 value the more potent the drug [33].

What is the difference between EC50 and IC50?

The EC50 is the concentration of a drug that gives half-maximal response. The IC50 is the concentration of an inhibitor where the response (or binding) is reduced by half.

What is the difference between LC50 and EC50?

Terminology: EC50(50\% Effective Concentration) The Lethal Concentration 50 (LC50) is the concentration of a substance in water causing a death (50\% of the tested population) to aquatic life. The EC50 and the LC50 are often used in ecotoxicology as an indicator of the toxicity of a compound to the environment.

How is IC50 calculated from MTT assay?

IC50 values were calculated by linear approximation regression of the percentage survival versus the drug concentration. Cell survival rate (\%)=(a-c)/(b-c) x100 (a=absorbance at each concentration of the anticancer reagent, b=absorbance at 0 µM of the anticancer reagent, and c=absorbance of the blank).

Is a high IC50 good?

IC50 is the dose of the substance which inhibits the proliferation of half of the cells. It does not necessarily need to kill the cells. If you find high IC50, this means that the cell line is more resistant to drug1 than drug 2. Then, you can suspect from other cell deaths, too.

Is potency equal to efficacy?

Efficacy is the ability of a drug to elicit a physiologic response when it interacts with a receptor. Potency is the amount of drug needed to produce a certain response.
